摘要 目的:研究雷公藤甲素对瘢痕成纤维细胞增殖的影响,为临床增生性瘢痕的防治提供科学的实验依据。方法:采用MTT检测雷公藤甲素对体外培养人瘢痕成纤维细胞增殖的影响,LDH检测细胞毒性。Giemsa染色观察细胞形态,RT-PCR检测TGF-β1的表达。结果:10、20和40μg/ml的雷公藤甲素对体外培养的瘢痕成纤维细胞均有抑制效应;雷公藤甲素作用24h后,可使瘢痕成纤维细胞的突出变短、变少,胞核、胞质浓缩;与对照组比较,雷公藤甲素可降低TGF-β1mRNA水平的表达。结论:雷公藤甲素对瘢痕成纤维细胞的增殖有抑制作用;雷公藤甲素降低TGF-β1mRNA的表达。 Objective :To investigate the effect of triptolide on proliferation of fibroblasts.Methods :Cell viability were determined by MTT assay.Cell toxicity was determined by lactate dehydrogenase assay.Cell morphology was determined by Giemsa staining.The human mRNA levels of TGF-β1 in fibroblasts were determined by RT-PCR.Results:Compared with control groups, triptolide caused statistically significant inhibition of cell viability(P〈0.01)without cell toxicity(P〉 0.05)Triptolide caused statistically significant decreases in the level of TGF-β1 compared with control groups (P〈0.01).Conclusion: 1.Triptolide can inhibit proliferation of the fibroblasts in vitro.2.Triptolide caused decreases production of TGF-β1 mRNA in fibroblasts in vitro.
