摘要
以分离到的一株德氏乳杆菌(Lactobacillus delbruckii) 1-1为出发菌株,经紫外线、硫酸二乙酯、亚硝酸钠等诱变剂处理,筛选出一株产乳酸较高的突变株ZL-513,乳酸钙产量由5.8%提高至9.4%。以正交试验为基础对发酵培养基进行优化,采用优化后的培养基发酵 3 d,残糖降至 0.1%以下,乳酸钙产量达 15%。比优化前产酸提高 60%,缩短发酵周期 1d。以质量分数 8%~15%的玉米粉为原料进行发酵,乳酸钙产量分别为10%~18.5%。
Parent strain Lactobacillus delbruckii 1-1 which accumulated 5.8% of calcium lactate was mutated by UV-ray, DES and NaNO2, and high lactic acid-producing mutant ZL-513 which accumulated 9.4% of calcium lactate was isolated. Fermentative medium was optimized on the basis of orthogonal design. Under the optimal medium the accumulation of calcium lactate reached 15% after 3 days' fermentation and the remnant glucose was below 0.1%. This medium not only increased 60% of the yield of the lactic acid but also shortened the fermentative cycle by one day compared with the original medium. The optimal medium consists of (%): glucose 10, yeast extract 1 .0, peptone 2. 0, K2HPO4 0. 06, MgSO4·7H2O 0.02, NaCl 0.08, beef extract 0.4. When using 8%, 10%, 12% and 15% of corn starch as raw fermentation materials, the corresponding yields of calcium lactate were 10%, 12%, 15% and 18 .5% respectively.
