摘要
目的:用表面增强激光解析离子化飞行时间质谱(SELDI-TOF-MS)分析肝内胆管癌和健康者血清蛋白指纹图谱的变化,建立能鉴别肝内胆管癌和健康者血清标志物的诊断模型。方法:收集33例肝内胆管癌和31例健康者的血清,用SELDI-TOF-MS及配套H50(疏水芯片)蛋白质芯片检测蛋白指纹的表达。用Biomarker Patterns Software分析软件进行数据处理,建立区分肝内胆管癌和健康者的蛋白质指纹图谱表达模型。结果:在分子量2000~20000范围内,共检测到127个有差异的蛋白峰,其中10个差异有显著性(P〈0.01),建立了由2个差异蛋白组成的肝内胆管癌诊断模型,其诊断灵敏度为96.9%(32/33),特异性为96.7(30/31)。结论:用SELDI-TOF-MS技术初步建立的区分肝内胆管癌和健康者的蛋白指纹表达模型可以区分肝内胆管癌和健康者,可能为肝内胆管癌的诊断提供新的手段。
Objective:To analyze the spectrometric serum protein profiling of intrahepatic cholangiocarcinoma(ICC) and healthy controls by surface-enhanced laser desorption/time-of-flight mass spectrometry(SELDI-TOF-MS) to establish serum marker pattern for the diagnosis of ICC.Methods:The serum samples in 33 cases of ICC and in 31cases of healthy controls on H50 proteinchip were collected,the spectrometric protein profiling was detected by SELDI-TOF-MS,the data were analyzed by biomarker patterns software provided by ciphergen corp.A primary diagnosis model of ICC was set up.Results:127 protein peaks were detected at the molecular range of 2000~20000Da,among which 10 ones were significantly different between ICC and controls(P0.01).A diagnostic pattern consisting of 2 protein peaks was established with sensitivity of 96.9%(32/33),specificity 96.7(30/31).Conclusion:It is successful to develop and evaluate the different spectrometric protein profiling patterns of ICC and healthy control by SELDI-TOF-MS.This provides possibly a new method to diagnosis of ICC.
