摘要
目的:探讨p53、p21、p185蛋白表达及p53、MDM2基因与横纹肌肉瘤(RMS)分化和预后的关系,并对几种检测方法进行了比较。方法:应用免疫组化方法检测RMSp53、p21、p185蛋白表达,银染PCRSSCP检测RMSp53的突变,原位杂交检测RMSp53和MDM2基因。结果:p53、p21、p185蛋白在RMS的阳性率分别为672%(41/61)、68(34/50)、60%(30/50);检测30例RMS,p53基因7例发生点突变原位杂交检测RMS的p53、MDM2基因突变的检出率分别为667%和774%;上述结果均与年龄、性别、肿瘤组织类型无关,而与肿瘤分化转移和预后有关。结论:p53、p21、MDM2蛋白表达及癌基因检测均有助于判断肿瘤恶性程度和肿瘤预后,上述3种方法比较,免疫组化与原位杂交检测的阳性结果基本相符,提示免疫组化方法同样能反映肿瘤恶性程度和预后,是一种简便可行的检测方法。
Objective:To observe the correlation between expression of oncogene protein products p53、p21、p185 and P53、MDM2 genes and differentiation and prognosis of rhabdomyosarcoma(RMS) and to compare these detection methods. Methods:Expression of oncogene protein products p53、p21、p185 in RMS were detected by immunohistochemical method.p53 mutation in RMS was detected by PCR SSCP method, p53 and MDM2 genes were detected by in situ hybridization.Result:The positive rates for expression of protein products of oncogene p53、p21、p185 were 67 2% 68% and 60% respectively.There were 7 cases p53 gene mutation in 30 cases of RMS.The positive rates for p53 and MDM2 genes were 66 7% and 77 4%,respectively.regardless of age,sex or RMS histological type,but they were related to the degree of RMS differentiation, metastasis and prognosis.Conclusion:The detection of expression of protein products of oncogen p53、p21、MDM2 and their genes has practical value for a judging maligant degree of tumor and prognosis.Comparing three above mentioned methods each other,showed that the detection result of immunohistcchemistry and in situ hybridization was conformable,this method is able to reflect maligant degree of tumor and prognosis.It is a simple and convenient method.
