摘要
采用体外培养人胚关节软骨细胞的方法,研究黄嘌呤-黄嘌呤氧化酶系产生的自由基对软骨细胞生长的影响。倒置显微镜下,软骨细胞生长缓慢、形态异常、没有集落形成。通过测定软骨细胞DNA、蛋白质、蛋白多糖及胶原的含量变化,发现自由基可显著抑制DNA、蛋白质及蛋白多糖的合成。采用流式细胞仪对软骨细胞周期进行分析,发现DNA合成后期(G2+M期)细胞数显著增加。显然,自由基通过对软骨细胞DNA的损伤,从而影响细胞分裂增殖,使细胞生长异常,破裂死亡,是自由基损伤软骨细胞的重要途径之一。
hondrocytes in monolayer culture were used to observe the damaging effects of oxygen free radicals generated by xanthine oxidase and xanthine. Under the microscope, the chondrocytes were seen grew slowly and there was no clone formed. By measuring the syntheses of DNA protein proteoglycan and collagen, the authors found out that free oxygen radicals could significantly inhibit the syntheses of DNA protein and proteoglycan but not collagen. Flow cytometry analysis revealed a perturbation in cell cycle progression lead to a significant increase in the proportion of cells in G 2+M phase. Apparently, it is one of important paths that free radicals inhibit cytodieresis , chondrocyte growth and eventually cause death of the cultured cells.
