摘要
目的用分子生物学方法进一步证明革螨在传播两型HFRS中的作用。方法将叮刺感染姬鼠型(76—118株)HFRSV乳鼠后第10天厩真厉螨270只和家鼠型(UR株)HFRSV后第10天厩真后螨200余只,分别制成组织悬液接种正常乳鼠,用Dig标记HFRSVCDNA探针原位分子杂交检测接种后第9天鼠肺;将叮刺感染乳鼠后螨在适宜条件下饲养至100天以上,分别叮刺4~5日龄正常乳鼠,于第3天取鼠肺冰冻切片,经Dig标记HFRSVCDNA探针原位分子杂交检测。结果叮刺76一118株病毒感染乳鼠螨悬液接种4只乳鼠,3只阳性,UR株接种6只,全部阳性;76—118株组饲养至132天,UR株组102天的螨叮刺正常乳鼠后,也从鼠肺中检出病毒RNA。结论首次用分子生物学方法证明该螨可通过叮刺获得姬鼠型和家鼠型HFRSV,螨群体不但维持76—118株病毒132天以上、UR株102天以上,而且均能经叮刘传播,说明此螨能作为姬鼠型和家鼠型HFRSV的适宜传播媒介和贮存宿主,这一发现为我国的HFRS疫区保存和演变提供了一定的实验依据,有流行病学意义;原位分子杂交可缩短用敏感动物分离革螨体内HFRSV的时间,可用于革螨等媒介昆虫传HFRSV的实验研究和流行病学调查。
Aim To further demonstrate the role of gamasid mites(Eulaepas stabularis)in the transmission of HFRS V,Hantaan and Seoul,using methods of molecular biology' Methos 2 or 3-day-old sucking mice were inoculated with tissue suspension of the 10th day mites after biting a suckling mouse infected with Hantaan(76 118) and Seoul(UR)strains respectively. At 9 days post -inoculation,detection of the mice lung was made with in situ hybridization of dig-labelled HFRSV M,S cDNA probes;some of the mites were reared to more than 100 days,then let the mites bite 4 or 5 day old suckling mice respectively. In 3 days from post -biting,detection of the mice lung was made with in situ hybridization of dig-labelled HFRS V M,S cDNA probes. Results The virus RNA was detected in lungs of 3 of 4 suckling mice of Hantaan group after inoculation of the tissue suspension of the mites,and all 6 mice for Seoul group respectively' The virus RNA was also detected in lungs of the mice bit by the mites. Conclusion The results confirmed that E .stabularis was sensitive to both Hantaan and Seoul strains of HFRSV and could keep the ability to transmit not only Hantaan but also Seoul strains for at least 132 days and 102 days respectively,and serves as a vector and reservoir of the two type viruses. The similar report has not been seen as yet. These findings provide a valuable evidence for keeping and changing of HFRS epidemic distTicts in China. Jn situ hybridization may short the time of isolation and detection of HFRSV from a sensitive animal,it can be a sensitive,specific and new rapid etiologic test method to be used for laboratorial and epidemiological investigation.
