氨磷汀对顺铂所致大鼠肾毒性的防护作用 被引量:1

摘要 目的研究氨磷汀对顺铂所致肾损害的防护作用,并探讨其可能机制。方法随机将36只大鼠分成3组,即对照组(氯化钠注射液4ml)、顺铂组(0.006mg/ml,4ml)和氨磷汀组(0.2mg/ml+0.006mg/ml;4ml)。分别测定体质量、肾脏脏器系数、血清生化指标、尿蛋白含量及N-乙酰β-D-氨基葡萄糖苷酶、碱性磷酸酶、r-谷氨酰转肽酶活性,并做肾组织病理学检查。结果氨磷汀组大鼠体质量较对照组低,但显著高于顺铂组。顺铂组大鼠的血清总蛋白、白蛋白均明显低于对照组,而尿素氮、肌酐均明显高于对照组,氨磷汀组与顺铂组相比有明显改善,更接近正常大鼠。氨磷汀组肾脏脏器系数较顺铂组明显下降,肾脏病理学检查显示氨磷汀可明显改善顺铂引起的肾小球充血、肾小管浊肿及管型的病理改变。结论氨磷汀对顺铂的肾毒性损伤有保护作用。 Objective To investigate the protective effect of amifostine on cisplatin -induced nephrotoxicity in rats and plausible mechanism. Methods Thirty-six Sprague-Dawley rats were randomly divided into three-groups: control group (saline solution 4 ml), cisplatin group(cisplatin 0.006 mg/ml,4 ml) and amifostine group(amifostine 0.2 mg/ml +cisplatin 0.006 mg/ml;4 ml). Body weight and organ coefficient of kidneys, serum biochemical indices, urine protein content and activity of N-acetyl-β-D-glocosaminidase,alkaline phosphatase,r-glutamyl transpeptidase were measured. Renal histopathological lesions were examined by light microscopy . Results Body weight in amifostine group was lower than that in control group but higher than that in cisplatin group. The serum levels of total protein and albumin were markedly lower but those of blood urea nitrogen and creatine higher in cisplatin group than those in control group.The above indexes were improved in amifostine group as compared with those in cisplatin group and approached the normal level. The organ coefficients of kidneys were decreased more obviously in amifostine group than those in cisplatin group. Pathological examination of the kidney showed that amifostine could relieve congestion of the glomerulus of the kidney, the cloudy swelling in renal tubules and pathological change of the tubules induced by cisplatin. Conclusion Amifostine has protective effect on renal toxicity induced by cisplatin.
