摘要
利用微卫星(SSR)标记技术寻找与桑树发芽率和节间距性状基因紧密连锁的分子标记,可为高产桑树品种的选育提供分子遗传学方面的依据。用10对SSR引物对172份桑树品种(品系)的基因组DNA进行扩增,结果在获得的67个标记中有66个具有多态性,多态性比率为98.51%。分析供试桑品种(品系)基因组SSR标记与桑树发芽率、节间距性状的关联性发现:SSR标记mulstr4-3、mulstr6-4、mlstr6-6、ss19-1与桑树的发芽率呈显著性相关,相关系数分别为0.209、-0.215、-0.208、0.226;SSR标记mulstr3-3、mulstr4-1、ss04-1、ss04-3、ss18-1与桑树枝条的节间距呈显著性相关,相关系数分别为-0.255、0.424、-0.248、0.216、0.229,其中标记mulstr4-1可能与控制节间距性状的基因相连锁。
In this paper, simple sequence repeat (SSR) marker technology was used to look for molecular markers closely linked to mulberry traits of budding rate and internodal distance, so as to provide molecular genetic basis for breeding high-yield mulberry. 10 pairs of SSR primers were used to amplify the whole genomes of 172 mulberry varieties (strains). The results demonstrated that, among the 67 DNA bands amplified, 66 bands showed polymorphism. The polymorphic rate reached 98.51%. Correlation analyses between genomic SSR markers and traits of budding rate and internodal distance of the tested mulberry varieties (strains) indicated that. SSR markers mulstr4-3, mulstr6-4, mlstr6-6 and ss19-1 were significantly correlated with the budding rate, with a correlation coefficient of 0. 209, -0. 215, -0. 208 and 0. 226 respectively. SSR markers mulstr3-3, mulstr4-1, ss04-1, ss04-3 and ss18-1 were significantly correlated with internodal distance, with a correlation coefficient of -0. 255, 0. 424, -0. 248, 0. 216 and 0. 229 respectively. Among these SSR markers, mulstr4-1 is probably linked to genes that control the internodal distance.
