活血利水法对外伤性增生性玻璃体视网膜病变模型兔眼IGF-1表达的影响 被引量:3

Effects of Method of Activating Blood and Diuresis on Expression of IGF-1 of Rabbits' Traumatic Proliferative Vitreoretinopathy Model

摘要 目的:观察活血利水之散血明目片对外伤性增生性玻璃体视网膜病变(traumatic Proliferrative Vitreo-retinopathy,tPVR)增殖膜(ERM)中胰岛素样生长因子-1(insulinlike growth factor-1,IGF-1)表达的影响。方法:将40只家兔随机抽取32只,造成外伤性PVR模型,随机分成活血利水组、活血化瘀组、利水明目组、模型组,另8只为空白组。连续灌胃28天后,观察眼底PVR分级情况,免疫组织化学方法检测胰岛素样生长因子-1(IGF-1)在各组的表现及病理组织学改变。结果:在活血利水组,ERM中胰岛素样生长因子-1(IGF-1)阳性程度低于模型组,差异有非常显著性(P<0.01)。活血化瘀组与利水明目组也能降低IGF-1的阳性程度,与模型组相比有显著性意义(P<0.05)。活血利水组IGF-1阳性程度低于活血化瘀和利水明目两个治疗组,有显著性差异(P<0.05)。结论:活血利水法能通过抑制玻璃体腔中胰岛素样生长因子-1(IGF-1)的作用,从而抑制增殖细胞的过度增生,防治PVR形成和发展。 Objective: To investigate the effects of SanXue MingMu Tablets (SXMMT) of activating blood and diuresis on expression of insulinlike growth factor - 1 ( IGF - 1 ) in epiretinal membrane (ERM) of traumatic proliferative vitreo- retinopathy. Methods :32 rabbits were selected randomly from 40 rabbits and made to traumatic proliferative vitreoretinopa- thy models, then divided into four groups : the first group was treated with activating blood and diuresis group, the second treated with activating blood and removing stasis group, the third treated with activating diuresis and brightening eye group and the model group. The rest 8 were formed the blank control group. After the rabbits had been administrated intragas- trieally for 28 days, we observed the classification of PYR and expressions and pathologic changes of IGF - 1 measured by immunohistochemistry in each group. Results : Compared with model group , the positive level of IGF - 1 was decreased significantly in activating blood and diuresis group (P 〈 0.01 ). Compared with model group, the positive level of IGF - 1 decreased much in activating blood and removing stasis group and activating diuresis and brightening eye group( P 〈 0.05). And the level of IGF - 1 in activating blood and diuresis group were lower than those in activating blood and remo- ving stasis group and activating diuresis and brightening eye group. Conclusions:Method of activating blood and dieresis could inhibit the expression of IGF - 1 of vitreous cavity as to inhibit proliferative cell~ hyperplasy and prevent and cure the formation and development of traumatic PVR.
