摘要
用作者等所构建的重组腺病毒AdCMVgdnf直接感染多种细胞系和大鼠中脑原代神经元细胞,多重感染复数为20PFU/ml。采用免疫组织化学染色方法检测GDNF表达,观察所构建的GDNF重组腺病毒在各种细胞中介导GDNF基因转移和基因表达的能力。结果表明在一系列神经细胞或非神经细胞中,GDNF重组腺病毒均可介导GDNF基因的高效转移和表达。提示GDNF重组腺病毒可作为帕金森氏病基因治疗的高效基因转移载体。
A number of cell strains were infected directly by recombinant adenovirus of AdCMVgdnf at a multiplicity of infec tion of 20, and the immunocytochemistry method was employed to examine the abilities that the recombinant adenovirus of AdCMVgdnf we constructed mediates GDNF gene transfer and expression in these infected cells. The results showed that the recombinant adenovirus of AdCMVgdnf can mediate gene transfer and expression of GDNF in all of the examined cells with high efficiency. It is concluded that the recombinant adenovirus of AdCMVgdnf might serve as an ideal viral vector system for gene transfer and gene therapy for Parkinson's disease.
