摘要
采用体内裸DNA基因治疗帕金森病(PD)取得显著疗效,将酪氨酸羟比酶(TH)基冈表达质粒与Lipofectin形成的复合物立体定位注射于PD模型鼠纹状体.显著改善了PD鼠的不对付旋转行为.
In vivo naked DNA gene transfer method wasused in gene therapy of Parkinson's disease(PD). The complex of rat tyrosine hydroxy-lase (TH) expression plasmid and Lipofectinwas injected stereotaxically into striatum of PDrat model. The asymmetric rotational behaviorwas reduced substantially and quickly. On thethird day after injection, drug-induced rotationdecreased 50% compared with pretreatmentscores. Immunohistochemical staining showedTH-positive nerve cells in striatum of injectionside, which indicated that TH gene was up-taked and expressed by nerve cells. These pre-liminary results have general implications forthe application of naked DNA transfer tech-nique in gene therapy of human neurologicaldisease and specific implications for PD.
