摘要
从家蚕品种C108和大造后部丝腺提取的基因组DNA,用PstⅠ酶解后与自己设计的人工接头相连,并用与人工接头序列相匹配的选择性引物进行选择性PCR扩增(selectiveamplification).扩增产物经琼脂糖电泳检测显示,用SADF法在两品种中能检测到稳定的DNA多态性片段.
Genomic DNA extracted from the posterior silk glands of two silkworm varieties,C108 and Dazao were digested with Pst Ⅰ and ligated with synthetic adapter.Selective amplification of DNA fragments (SADF) was performed using suitable primers which annealed to adapter regions.Agarose gel electrophoresis shown that DNA polymorphic fragments were appeared for both C108 and Dazao.
