影响油菜子叶外植体不定芽高频率再生的因素 被引量:34

FACTORS AFFECTING SHOOT REGENERATION AT HIGH FREQUENCY FROM IN VITRO CULTURE COTYLEDON OF BRASSICA NAPUS AND B.CAMPESTRIS

摘要 采用7个甘蓝型和2个白菜型油菜品种(系)研究了影响子叶外植体芽再生的一些因素和不同基因型的子叶外植体的离体培养反应和芽再生能力。结果表明,苗龄4d的幼苗子叶于含2,4-D0.5-1.2mg/L和6-BA0.2mg/L的培养基培养(称预培养)2或6d后再转到分化培养基上培养,芽再生率提高34%-46%。分化培养基中附加AgNO3(6mg/L)可促进不定芽的形成和生长,而丝氨酸和谷氨酰胺则没有表现出有益效应。在同一培养条件下,甘蓝型油菜子叶比白菜型再生能力强。在甘蓝型油菜基因型间,子叶再生能力差异很大。 The cotyledon explants of 7 Brassica napus and 2 B.campestris varieties (lines) were cultured and some of factors affecting shoot regeneration at high frequency were also studies.Results showed that the frequencies of shoot regeneration of cotyledons explants from 4 d old in vitro grown seedlings were markedly increased from 34% to 46% via a preculture procedure of 2 days period using the media containing the hormone combinations of 2,4 D 0.5-1.2 g/L and 6 BA 0.2 mg/L.AgNO 3(6 mg/L) added in regeneration media was advantageous for the formation of adventitions shoots whereas L serine and L glutamine was not good action on shoot formation.Under same culture condition,the shoot regeneration rate of B.napus and B.campestris was 28.85%-90.57% and 24.32%-27.78%,respectively.This indicated that the shoot regeneration capacity of interspecies and inter varieties (lines) was greatly different.
