重组人α-肿瘤坏死因子对移植至裸鼠后的增生性瘢痕的影响 被引量:1

摘要 目的:观察重组人α-肿瘤坏死因子(rhuTNF-α)对烧伤后增生性瘢痕组织胶原含量及组织形态学的影响。方法:12只BALB/c裸鼠,于背部体表移植人体烧伤后增生性瘢痕组织(2cm×2cm)成活后,随机分为试验组及对照组。试验组在增生性瘢痕组织内注射生理盐水溶解的rhuTNF-α0.1ml(500ng/ml),每周3次,对照组同法注射生理盐水,3周后测定瘢痕组织中胶原、羟脯氨酸及酸性粘多糖含量,同时观察组织形态学变化。结果:局部注射rhuTNF-α后,增生性瘢痕组织中胶原、羟脯氨酸及酸性粘多糖含量显著降低。增生性瘢痕组织变平、缩小,胶原结节明显减少或消失。成纤维细胞粗面内质网显著减少,无扩张。结论:rhuTNF-α能显著减少增生性瘢痕组织中胶原沉积,在增生性瘢痕治疗中可能有潜在应用价值。 Objective: To investigate the effects of recombinant human tumor necrosis factor α (rhuTNF α) on the collagen content in and histomorphology of postburn hypertrophic scar tissues. Methods: Twelve nude mice with human postburn hypertrophic scar tissues (2 cm×2 cm) transplanted on to their back and survived were randomized into the experimental group (EG) and the control group (CG). For animals in EG, 0.1 ml (500 g/ml) rhuTNF α dissolved in normal saline was injected into the hypertrophic scar tissues for 3 times a week. For those in CG, only normal saline was injected in the same manner. The contents of collagen, hydroxyproline and acidic mucopolysaccharide in and the histomorphological changes of the hypertrophic scar tissues were determined 3 weeks later. Results: As compared with CG, in EG the contents of collagen, hydroxyproline and acidic mucopolysaccride were significantly decreased. After the injection of rhuTNF α, the hypertrophic scar tissues became flatter and smaller. Collagenous nodules in hypertrophic scar tissues drastically decreased or disappeared. The number of rough endoplasmic reticulum in fibroblasts was markedly reduced and no dilatation was seen. Conclusion: rhuTNF α can remarkably reduce collagen deposition in hypertrophic scar tissues, which suggests that rhuTNF α may be of potential value in treating hypertrophic scars.
