大鼠局灶性脑缺血对大脑皮层Bax,Bcl-2基因表达的影响 被引量:6

摘要 目的 :为探讨局灶性脑缺血对大鼠大脑皮质神经细胞Bax ,Bcl- 2基因表达的影响 ,方法 :采用免疫组织化学ABC法 ,检测了局灶性脑缺血后 6h大鼠大脑皮质Bax ,Bcl- 2的表达 .结果 :正常组及假手术组 ,Bax ,Bcl- 2的免疫阳性物主要分布于锥体细胞层 ,胞浆染色 .缺血 6h后 ,神经元Bax ,Bcl- 2的平均灰度分别为 73 1± 6 6和 10 7 7± 9 1,分别较对照组 84 7± 4 4和 91 3± 5 5减少和增加 (P <0 0 5 ) .表明缺血后Bax含量增加 ,Bcl- 2含量减少 .提示 :缺血致大脑皮质神经元凋亡可能与Bax ,Bcl- In order to explore the change of Bax, Bc1-2 expression in cerebral cortex at 6?h following ischemia, rats were performed local ischemia operation derived from middle cerebral artery block. Rats were sacrificed at 6?h following ischemia. The cerebral cortex was taken and sectioned into 20?μm frozen section. The sections were stained using antibody of Bax, Bc1-2 by ABC immunohistochemical technique. The gray degree of Bax,Bc1-2 was measured by imagine analysis system. Results: At 6h following ischemia, the gray degree of Bax, Bc1-2 in neurons of cortex in control is respectively 84.7±4.4, 91.3±5.5 and 73.1±6.6, 107.7±9.1. It showed that expression of Bax was upregulated and Bc1-2 expression was decreased following ischemia.
