白羊草幼穗在附加有2,4-D1.0mg/L、水解酪蛋白500或1000mg/L的N6或MS培养基中,在25±1℃,暗光培养条件下,两周时形成0.5cm2大小的愈伤组织块。愈伤组织的发生率为84%。培养3周后形成旺盛生长的愈伤组织。其分化是在含有NAA0.5mg/L、KT1.0mg/L的MS培养基上或者在附加有KT1.0mg/L、IAA0.4mg/L、BA1.0mg/L的N6培养基上2~4周后均可得到旺盛生长的分化植株,将该植株移至不含激素的N6培养基内,在光照下进行培养,形成绿色分化植株。3~4周后,将分化植株移入土壤中,植株生长良好。
Cultured in darkness and under the temperature condition of 25±1℃ for 2 weeks, the young spikelet of Bothriochloa ischaemum, laid on N 6 or MS medium containing 2,4 D (1.0 mg/L) and CH (500 or 1 000 mg/L), produced callus of 0.5 cm 2 in size and with a rate of occurrence as high as 84%. The callus became well grown in one week and then it was cultured into vigorous shoots in 2~4 more weeks on a new medium of MS+NAA (0.5 mg/L)+KT (1.0 mg/L) or N 6+KT (1.0 mg/L)+IAA (0 4 mg/L)+BA (1.0 mg/L). The formed shoots were then transferred onto a medium of N 6 without hormones, and, under light, they developed into green plants in 3~4 weeks. The plants were successfully transplanted into soil and grew well.
