环磷酰胺诱导少精子/无精子症大鼠模型所致睾丸、附睾bFGF的变化 被引量:11

Changes of bFGF in testis and epididymis of a rat model with oligoazoospermia/azoospermia induced by cyclophosphamide

摘要 目的探讨环磷酰胺(CP)对大鼠附睾中碱性成纤维细胞生长因子(bFGF)浓度和睾丸组织中bFGF表达的影响。方法 96只8周龄成年雄性SD大鼠,分6组,每组16只。第1~5组为实验组,分别给予CP10、20、40、80、100mg/kg·d^(-1);第6组为对照组,给予生理盐水2ml。胃管给药2周和4周后,处死大鼠,采用ELISA法对附睾组织洗脱液进行细胞因子bFGF定量检测;采用S-P免疫组化法检测上述模型睾丸组织细胞因子bFGF的表达。结果与对照组比较,10、20、40、80、100mg/kg·d^(-1)组2周后bFGF分别下降了28.5%、49.1%、52.8%、57.6%、59.4%,4周后bFGF分别下降了57.8%、66.3%、77.5%、83.0%、83.8%;随着CP处理时间的延长和剂量的增加,细胞因子bFGF在大鼠附睾中的浓度逐渐下降(P<0.05)。大鼠睾丸组织中细胞因子bFGF的表达随着CP处理时间的延长和剂量的增加而降低(P<0.05)。结论 CP在引起大鼠少精子/无精子症的同时,可以显著降低大鼠附睾、睾丸bFGF的含量和表达水平。bFGF的降低与CP诱导大鼠生精功能明显相关。 Objective To investigate the levels of bFGF in the epididymis and the expression of bFGF in the testis of adult male rat under the administration of cyctophosphamide. Methods Ninety-six male adult rats (8 weeks age) were equally divided into 6 groups. The rats in groups 1, group2, group3, group4 and group5 were treated with 10, 20, 40, 80 and 100 mg/kg·d-1 cyclophosphamide respectively. The remaining group was treated with normal saline as control. On 14 and 28 days after the administration of the drug (by gastric tuble), the treated rats were sacrificed. The level of bFGF was determined by ELISA. SP Immunohistochemistry was used to detect the expression of bFGF in testis tissues. Results Compared with that of the control, the level of bFGF in Groupl, 2, 3, 4, 5 decreased 28.5%, 49.1%, 52.8%, 57.6%, 59.4% respectively after 14 days, and 57.8%, 66.3%, 77.5%, 83.0%, 83.8% respectively after 28 days. Expression of bFGF in the epididymis of the rats was gradually reduced following with the increases of treatment time and doses (P〈0.05). In the mean time the expression of bFGF in the tissues of the testis of rats were also gradually reduced (P〈0.05). Conclusion In a rat model with oligoazorspermia/azoospermia induced by cyclophosphamide, the expression of bFGF in epididymis and testis tissue were significantly reduced. It indicated that decrease of bFGF might be contributed to spermatogeneous function of the testis.
