摘要
研究了悬浮培养昆虫细胞 Sf9时感染指数 (MOI)、感染时间和培养基营养状况对重组蛋白激酶原 (pro- UK)表达的影响。发现在转瓶培养中采用 MOI3、感染对数中期细胞所产生的重组蛋白的量最高 ,酶活可达 42 0 U/ml。在反应器中采用同样感染条件 ,重组蛋白的表达量可达530 U/ml。在转瓶培养中采取感染后换液处理 ,重组蛋白的表达可提高至 61 0
The effect of multiplicity of infection (MOI),infection time and medium nutrient on the recombinant protein(pro UK) expression was studied in Sf9 insect cell suspension culture.In spinner flask,MOI 3 and infection at middle exponential phase were suitable for the recombinant proteinexpression.The expression level was 420U/ml.In 2 L bioreactor,the expression level reached 530U/ml.The increase may be caused by the sufficient oxygen in bioreactor, and the medium replacement post infection made the expression level rise to 610U/ml in spinner flask.
