摘要
目的:本实验建立小鼠脑缺血模型,观察黄芪对小鼠脑缺血热休克蛋白70的表达影响。方法:将48只健康昆明小鼠随机分为三组:假手术组、缺血组、黄芪组。黄芪组腹腔内注射黄芪注射液2.5g/kg,缺血组和假手术组腹腔内注射等量生理盐水,连续7天后用微动脉夹夹闭小鼠双侧颈总动脉20min,手术后即刻断头取脑、冷冻,进行热休克蛋白70的定量分析。结果:与假手术组相比,缺血组热休克蛋白70的表达升高58%,黄芪组热休克蛋白70的表达升高32%;与缺血组相比,黄芪组热休克蛋白70的表达降低16.5%。结论:脑缺血敏感地诱导了热休克蛋白70的表达,而黄芪的应用明显地抑制了其表达。
Objective:This experiment establishes the mouse cerebral ischemia model to observe astragalus how to influence the expression of HSP 70. Methods: 48 health Kunming mice were divided into three groups at random: Sham group, ischemia group, astragalus group. Mice of astragalus group were injected astragalus inoculation fluid 2. 5g/kg into abdominal cavity, others were injected the isometric physiological saline ;Td later, all mice were blocked bilateral common carotid artery with artery clip for 20. then were taken the brain tissue, frozen and carried on the HSP70 quantitative analysis. Results: Compared with the sham group, the ischemia group HSP70 expressions increased 58% , the astragalus group HSP70 expressions increased 32% ; Compareed with the ischemia group, the astragalus group HSP70 expressions reduced 16.5%. Conclusion: The cerebral ischemia has induced the HSP70 expressions sensitively, but the astragalus application has suppressed its expression obviously.
