摘要
用SD大鼠肾皮质总RNA克隆的RAP-cDNA,经原核融合蛋白表达系统获得纯化的受体相关蛋白(RAP),由此制备特异的兔抗血清。分别提取大鼠各组织蛋白,经蛋白质聚丙酰胺凝胶电泳(SDS-PAGE)分离后,转移至硝酸纤维素膜,用抗RAP抗血清进行蛋白质印迹分析(WesternBlotAnalysis)。结果显示,RAP在大鼠肝、胰、肾皮质、肾上腺、大网膜、大脑、小脑、骨胳肌等组织内均有不同程度的表达,肺和脾脏内未见。推测RAP参与许多组织器官的代谢过程,尤其可能是与脂蛋白代谢有关的重要物质。
The RAP-cDNA obtained by RT-PCR was cloned into PGEX expression vector. The purified RAP fusion protein from prokaryotic system is injected into rabbit. Finally we get antiserum to RAP. After electrophoresis of lysate from different tissues in SDS-PAGE and transferring into the NC membrane, we used the antiserum to RAP in Western Blot Analysis. The results reveals that RAP is expressed highly in liver, kidney, adrenal gland and fat tissue, and also in brain, cerebellum and muscle. It was not found in lungs and spleen. These suggest that RAP may take part in the process of lipid metabolism in many tissues for RAP is chaperone of LDL-receptor family.
