摘要
目的对肥大细胞在实验性肝纤维化过程中形态与功能变化进行动态观察并初步探讨其作用。方法通过组织化学、电镜技术、图像分析及免疫组织化学等方法对肥大细胞数量、组化性质及超微结构改变、肝纤维化程度进行动态观察,并对TNFα的分布进行定位研究。采用荧光生化测定肝脏组胺含量,酶联免疫法测定血浆TNFα浓度。结果随肝纤维化程度逐渐加重,各组肥大细胞数目和肝组织组胺浓度均逐渐增高,与正常对照组相比增高显著(P<005),且二者均与血浆TNFα浓度变化正相关。结论肥大细胞在肝纤维化的发生发展中起重要作用,其作用通过功能与结构变化而实现。
Objective To investigate the morphologic and functional changes of mast cells (MCs) and go further into the role of MCs in the experimental liver fibrosis. Methods By using rat liver fibrosis model induced with carbon tetachloride and ditetic factors histochemical, immunohistochemical and electromicroscope techniques were employed to observe the dynamic changes of MCs numbers, histochemical charateristics, ultrastructure and tumour necrosis factor (TNFα) distribution in the liver. Liver histamine contents and plasma TNFα concentrations were respectively determined with fluorescent biochemical and ELISA means. Results MCs numbers and hepatic histamine contents of all liver fibrosis groups both tended to increase and were significantly higher than those of control group (P<0 05). There were positive correlations existing between plasma TNFα and hepatic histamine contents (r=0 8329, P<0 001), plasma TNFα and MCs population (r=0 8121, P<0 02). At 6th and 8th week of the liver fibrosis, it was found that MCs predominatly accumulated in the fibrous septa area and there was obvious degranulation. TNFα-positive cells located in both sinusoidals and fibrous septa area of the fibrotic liver, but only in the sinusoidals of normal rat liver. Conclusions MCs play an important role which relates to their functional and structual alteration in the liver fibrosis process. The alteration may be modulated by TNFα.
