摘要
目的探索简单、经济易行的大容量血样无菌分离外周血单个核细胞(PBMC)的方法。方法同时用新方法和经典的聚蔗糖-泛影葡胺密度梯度离心法分离同一份血样。配对t检验分析比较两种方法获取PBMC的数量。新方法采用先离心抗凝血,取出血浆与红细胞交界层,再用经典分离法分离交界层获得PBMC,余留抗凝血再次离心取出交界层并分离获取PBMC。结果新方法得到的PBMC总数不低于经典分离法(P>0.05)。结论该法能简单安全地从大量血样中无菌分离PBMC。
Objective To explore an economical and practical way for isolating PBMC from high-capacity peripheral blood with sterile operation. Methods PBMCs were isolated from the same peripheral blood sample by the new way and the classic way of Ficoll-Hya- que density gradient centrifugation respectively. The quantity of the PBMC isolated by the two different ways were analyzed by paired t- test. By the new way,the anticoagulated blood was centrifugated,then the boundary layer between plasma and erythrocytes was extract- ed, and finally PBMCs were isolated and gained from the boundary layer by the classic way. This process was repeated to get more PB- MC from the remnant blood. Results The PBMCs isolated through the new way were not less than that of the classic way(P 〉 0.05 ). Conclusion The new way is simple and easy, and can be used to isolate PBMCs from the high-capacity peripheral blood with sterile operation.
