摘要
以活性炭为载体固定化脂肪酶,研究了活性炭粒度和吸附条件对脂肪酶固定化的影响。结果表明,随着活性炭粒度减小,其吸附酶量和固定化酶活力增大,但吸附酶量过大会导致酶分子相互凝结,降低固定化酶的催化活性。当酶液质量浓度在0.01~0.05mg/mL时,提高酶液质量浓度可以提高载体吸附酶量和固定化酶活力。酶液pH和温度仅对固定化酶活力有影响,而对载体吸附酶量无影响。在pH7.0、30℃和150r/min振荡条件下,采用40目活性炭吸附0.05mg/mL酶液1h,制得的固定化脂肪酶活力达65U/g。
The effects of particle size and adsorption condition on lipase immobilized in active carbon were investigated in this paper. The smaller the particle sizes, the higher adsorption and immobilized lipase activity. The stronger lipase adsorption on active carbon might condensate the lipase resulting in lower lipase activity. An increase in lipase concentration from 0.01 to 0.05 mg/mL could improve the activity and absorption of immobilized lipase. The pH and temperature would affect the immobilized activity, but not adsorption. The immobilized lipase activity could reach 65 U/g when prepared by shaking the mixture of 0.05 mg/mL lipase solution at pH 7.0 and active carbon particles of 40 mesh at 150 r/min, 30 ℃ for 1 h.
