摘要
目的鱼腥草素钠(SH,清热解毒中药)对肾小球肾炎疗效和作用机制的实验研究。方法BALB/C小鼠随机分为4组:空白组、模型组、SH低、高剂量(60,120mg·kg-1)。建立肾小球肾炎模型,检测蛋白尿变化、肾脏组织学改变及肾脏组织中单核细胞趋化蛋白-1(MCP-1)表达的情况。结果小鼠给予SH后,蛋白尿较模型组显著减少;SH能改善肾脏组织的组织学情况,并能显著降低肾脏组织中MCP-1的表达(P<0.01)。结论SH治疗肾小球肾炎的作用机制可能是通过影响尿蛋白表达、肾脏组织学结构及MCP-1的蛋白表达而发挥作用。
Objective To investigate the efficacy and possible mechanism of sodium houttuygonate (SH) on the glomerulonephritis in mice. Methods A total of 48 BALB/C mice were randomly divided into four groups, including normal group, model group, low and high dose (60,120 mg·kg-1) SH groups. BALB/C mice were injected through the caudal vein with cationic Bovine Serum Albumin(C -BSA) to create glomerulonephritis models. Mice urine protein was analyzed by the test thrips very week. Morphological of the kidney was observated by the light microscope. The expression of monocyte chemoattractant protein 1 ( MCP -1 ) was detected by the immunohistochemistry. Result Compared with the normal group, the expression of the urine protein was significantly increased in the model group. And the expression of urine protein in the tiral groups was significantly decreased compared with the model group. SH can cured the morphological character of the kidey induced by the C - BSA. The expression of MCP - 1 protein was significantly decreased in the groups after SH used compared with the model group. Conclution SH can protect against C - BSA induced glomerulo- nephrits in BALB/C mice through the suppression of the urine protein, morphological character and MCP - 1 expressions. Therefore, SH had an effective action in glomerulonephrits induced by the C - BSA, and the likely mechanism can account to influence the urine protein, morphological character and MCP - 1 protein in the mice.
