摘要
目的:研究Smad4及Smad7在不同胃组织中的表达及作用。方法:病理学证实的胃癌根治术切除的胃癌组织标本78例,胃癌旁组织78例,采用SP免疫组织化学染色技术,检测Smad4及Smad7在不同胃组织中的表达。结果:①Smad4蛋白在胃癌旁组织中的阳性表达率为69.23%,在胃癌组织中的阳性表达率为44.87%(P<0.01);随着胃癌分化程度的降低其阳性表达率降低;有淋巴结转移者阳性表达率为23.25%,无淋巴结转移者的阳性表达率为71.43%,差别亦有统计学意义(P<0.01)。②Smad7蛋白在胃癌旁组织中的阳性表达率为17.95%,在胃癌组织中的阳性表达率为89.74%(P<0.01);随着胃癌分化程度的降低其阳性表达率升高;有淋巴结转移者阳性表达率为97.67%,无淋巴结转移者的阳性表达率为80.00%,有淋巴结转移者阳性表达率高于无淋巴结转移者(P<0.05)。结论:①Smad4蛋白在胃癌组织中低表达,与癌旁组织比较差异有统计学意义(P<0.01),其表达的阳性率与胃癌的分化程度及转移有关。②Smad7蛋白在胃癌组织中高表达,与癌旁组织比较差异有统计学意义(P<0.01),其表达的阳性率与胃癌的分化程度及转移有关。
Objective: To investigate the expression of Smad4 and Smad7 in different gastric carcinoma tissues, and to explore their effect on gastric cancer. Methods: The expression of Smad4 and Smad7 was examined by using immunohistochemistry (SP method) in different gastric tissues,including 78 cases of pathologically confirmed gastric carcinomas and 78 cases of adjacent non-cancerous mucosa. Results:(~The positive rate of Smad4 expression was 69.23% in lateral spherical aberration tissues of gastric cancer, and 44.87% in gastric cancer tissues(P〈0.01 ). The positive rate of Smad4 expression was descended as the differentiated degree of gastric cancer was descended. There was also a significant difference in the Smad4 expression between lymph node metastasis positive and lymph node metastasis negative tumors(23.25% Vs 71.43%, P〈0.01 ). (~YI'he positive rate of Smad7 expression was 17.95% in lateral spherical aberration of gastric cancer tissues, and 89.74% in gastric cancer tissues(P〈0.01 ). The positive rate of Smad7 expression was increased as the differentiated degree of gastric cancer was descended. There was also a significant difference in the Smad7 expression between lymph node metastasis positive and lymph node metastasis negative tumors (97.67% vs 80.0% ,P〈0.05). Conclusions: (1)The expression of Smad4 was low in gastric cancer tissue, Which was significantly difference that in the lateral spherical aberration of gastric cancer tissues (P〈0.01). The positive rate of Smad4 expression was related to the differentiated degree and metastasis of gastric cancer.(2)The expression of Smad7 was high in gastric cancer tissue, which was significantly difference that in the lateral spherical aberration of gastric cancer tissues(P〈0.01 ). The positive rate of Smad4 expression was related to differentiated degree and metastasis.
