摘要
在单因素试验的基础上,利用响应面分析方法优化荷叶中黄酮化合物的超声提取条件。利用中心组合设计研究乙醇浓度、液固比、超声时间、超声温度4个自变量对响应值黄酮得率的影响。用SAS9.0进行结果分析,响应面的典型分析可知稳定点是最大值,决定系数为0.9598。黄酮最佳超声提取工艺条件为:乙醇浓度61%,液固比33:1,超声提取时间30min,超声提取温度66℃,浸提次数2次,在此条件下黄酮得率为4.584%。
On the base of the single factor experiments,the optimum conditions for the ultrasonic extraction of flavonoids from lotus leaves were determined using response surface methodology (RSM).A central composite design was used to investigate the effects of four independent variables,namely ethanol concentration,the liquid to solid ratio,ultrasonic extraction time and ultrasonic extraction temperature on the response,yield of flavonoids.Data were analyzed using SAS9.0 software.Canonical analysis of surface responses revealed that the stationary surface was a maximum.The coefficient of determination was 0.959 8.The optimum ultrasonic extraction conditions were:ethanol concentration 61 %,the liquid to solid ratio 33:1,extraction time 30 min,extraction temperature 66 ℃,extracting two times.Under the optimum conditions,the extraction yield of flavonoids was 4.584 %.
