摘要
代谢组学(metabonomics)作为系统生物学的重要组成部分,已成为一项新的研究热点。代谢组学旨在对生物体液代谢物进行定性、定量研究。液相色谱和质谱联用(liquid chromatography-massspectrum,LC-MS)技术已应用于代谢组学研究。高效液相色谱和质谱联用(high-performance liquid chromatography-mass spectrometry,HPLC-MS)技术具有检测样本多样性、可重复定量分析、具足够灵敏度及选择性、分析快速且方便、可分析体液中结构复杂分子等优势。本文就高效液相色谱和质谱联用技术及其用于代谢组学研究的现状综述如下。
Metabonomics, an important branch of systems biology, has become a new hotspot. Metabonomics aims at the comprehensive and quantitative analysis of wide arrays of metabolites in biological samples. High-performance liquid chromatography-mass spectrometry (HPLC-MS) is now expanding into endogenous metabolite research. Its main advantages include wide dynamic range, reproducible quantitative analysis, high sensitivity and selectivity, rapidity and convenience and its ability to analyze molecules with extreme complexity in biofluids. Here, we review the strategy and application of high-performance liquid chromatography-mass spectrometry based metabonomics.
