杀菌剂对莲瓣兰种子原生地萌发胚培养中真菌污染控制的研究 被引量:5

Research on the Controlling of Fungicide on the Fungus Contamination in the Germination Embryo Culture of Cymbidium tortisepalum's Seeds in situ

摘要 [目的]研究不同杀菌剂对莲瓣兰杂交种子原生地共生萌发突破种皮的胚培养过程中真菌污染控制及胚培养成活情况的影响。[方法]以莲瓣兰杂交种子原生地共生萌发突破种皮的胚为外植体,分别用1.0g/L代森锰锌、1.0g/L多菌灵、1.0g/L甲霜灵和1.0mg/L培福朗预处理外植体5min,再按常规灭菌后将外植体接种到改良RM培养基中在适宜条件下培养,观察胚培养过程中真菌污染控制及胚培养成活情况。[结果]莲瓣兰杂交种子原生地共生萌发突破种皮的胚经1.0g/L代森锰锌和1.0mg/L培福朗预处理5min,胚培养过程中真菌污染率控制在10.0%以内,显著优于无处理的对照组(CK);代森锰锌处理胚培养成活率较低,胚培养成活率仅为67.5%,培福朗处理胚培养成活率显著高于代森锰锌,胚培养成活率高达84.8%。[结论]培福朗是莲瓣兰胚培养理想的杀菌剂。 [Objective]The research aimed to study the effects of different fungicides on the fungus contamination control and the embryo culture survival in the process of symbiotic germination embryo cultuture of hybrid seeds of Cymbidium tortisepalum in situ.[Method]Using the symbiotic germination embryo hybrid seeds of Cymbidium tortisepalum in situ as explants,the explants were pretreated with 1.0 g/ml Mancozeb,1.0 g/ml carbendazim,1.0 g/ml etalaxyl and 1.0 g/ml uazatine for 5 min respectively.After conventional sterilization,the explants were inoculated in improved RM culture medium and cultured under suitable conditions to observe the fungus contamination control and embryo survival ratio in the course of embryo culture.[Result]The fungus contamination percent of embryo form of Cymbidium tortisepalum's hybrid seeds in situ symbiotic germination was controlled within 10% by using 1.0 g/L Mancozeb and 1.0 mg/L uazatine for 5 minuts,significantly superior to control group.The survival ratio of embryo culture with Mancozeb was lower,only 67.5%.The survival ratio of embryo culture with uazatine was remarkably higher than Mancozeb and the survival ratio of embryo culture was 84.8%.[Conclusion]Uazatine was the perfect fungicide of Cymbidium tortisepalum's embryo culture.
