补肾调周法对多囊卵巢大鼠性激素和胰岛素样生长因子-1的影响 被引量:10

Effects of Method of Invigorating Kidney and Regulating Menstruation on Sex Hormone and Insulin-like Growth Factor-1 Levels in Rats with Polycystic Ovary

摘要 目的观察补肾调周法对造模大鼠性激素和胰岛素样生长因子-1(IGF-1)的影响。方法将60只大鼠随机分为6组,即空白组,模型组,补肾调周低、中、高剂量组,阳性对照组,每组各10只。除空白组外其余各组采用HCG+胰岛素(INS)建立多囊卵巢(Polycystic Ovary,PCO)动物模型。补肾调周法序贯治疗16 d后测定血清T、LH、E2、INS含量,免疫组化法检测胰岛素样生长因子-1(IGF-1)在卵巢组织的表达,并在光镜下观察卵巢组织形态。结果补肾调周法能显著降低PCO模型大鼠血清T,LH,E2,INS含量,减少IGF-1在卵巢组织的表达,增加卵巢颗粒细胞层数,增加黄体组织数量。结论补肾调周法可改善多囊卵巢综合征高胰岛素血症和高雄激素血症,且具有多途径、多环节、多靶点的特点。 Objective To observe the effects of method of invigorating kidney and regulating menstruation on sex hormone and insulin - like growth factor - 1 ( IGF - 1 ) levels in rats with polycystic ovary. Methods 60 rats were randomly divided into 6 groups (n = l0 ) : blank group, model group, low dose of invigorating kidney group, middle dose of invigorating kidney group, high dose of invigorating kidney group, masculine control group. In addition to the blank group, the other groups were induced as PCO model by insulin and HCG. After 16 days treatment with the method of invigorating kidney and regulating menstruation, the serum levels of testosterone ( T), luteinizing hormonl (LH) , estradiol ( E2 ) and insulin (INS) were determined, the expression of IGF - 1 in the ovary was detected with immunohistochemical method, and organization shape of owary was observed under the light microscope. Results The method of invigorating kidney and regulating menstruation could obviously reduce the serum levels of T, LH, E2, and INS content in PCO model rats, inhibit the expression of IGF - 1 in the ovary and increase the granular cell layer and the quantity of the yellow body organization of ovary. Conclusion The method of invigorating kidney and regulating menstruation may improve the hyperinsulinemia and hyperandrogenism of PCOS with the characteristics of multi - ways, multi - links and the multi - target.
