摘要
建立了高效液相色谱法测定小鼠脑中四种氨基酸类神经递质含量的方法。以2,4-二硝基氟苯(DNFB)为衍生化试剂,Diamonsil C18(4.6×200mm,5μm)色谱柱,乙腈和醋酸钠缓冲液为流动相,采用二元梯度洗脱,UV350nm检测。结果显示:在18min内四种氨基酸分离完全,在0.01~100ug·mL^(-1)范围内线性关系良好,Asp、Glu、Gly、GABA相关系数分别为0.998 5,0.997 6,0.994 3,0.992 4;日内变异系数分别为2.4%,3.8%,3.2%和3.5%;标准加入平均回收率分别为95.18%,94.70%,97 60%和95.90%。本方法快速、准确、简单、灵敏度高、重现性好,为临床检测及科研提供参考。
Established a method for the determination of amino acids neurotransmitter in rat brain by RPHPLC. The samples were pre-column derivated with DNFB,Diamonsil C18column(4.6×200mm,5μm),acetonitrile /acetate buffer solu-tion as mobile phase,gradientelution with UV 350nm detection.According to the study, amino acids were separated within 18 mins.The linear re-lation was well in the range from 0.01 to 100ug/ml,The correlation coefficients were:Asp0.9985,Glu0.9976,Gly 0.9943,GABA0.9924 respectively.The precisions were Asp2.4%,Glu3.8%,Gly 3.2%and GABA3.5%for interlay:The average recoveries of amino acid were Asp 95. 18%,Glu 94.70%,Gly 97.60%and GABA95.90%respectively.The method was speediness,accurate,simple, high sensible and well repetition,and supplied a reference for the clinic determination and scientific research.
