RP-HPLC法测定大鼠脑组织及脑脊液中四种氨基酸类神经递质的含量 被引量:10

摘要 目的建立大鼠脑组织及脑脊液中四种氨基酸类神经递质含量的RP-HPLC测定方法。方法以2,4-二硝基氟苯(DNFB)为衍生化试剂柱前衍生,色谱条件为大连依利特色谱柱Hypersil BDS C18(4.6×200mm,5μm),乙腈和醋酸钠缓冲液为流动相,采用梯度洗脱,UV360nm检测。结果在25min内四种氨基酸分离完全,在1~100ng.mL-1范围内线性关系良好(r≥0.9993)。结论本方法准确,简便,灵敏度高,分析时间短,适用于一般实验室氨基酸的检测。 Objective To establish a method for the determination of 4 amino acids neurotransmitter in rat brain tissue and cerebrespinal fluid by RP-HPLC. Methods The samples were pre-column derivated with DNFB, analyzed by a Yilito Hypersil BDS C18 column (4.6×200mm,5μm)as stationary phase , acetonitrile/acetate buffer solution as mobile phase, gradient elution with UV 360 nm detection. Results 4 amino acids were separated within 25 min. The linear relation was well in the range from 1ng·mL^-1 to 100ng·mL^-1(r≥0.9993). Couclusions The method is accurate, simple, high sensible and short analytical time. It can be used successfully in common laboratory.
