HPLC测定感冒灵颗粒中对乙酰氨基酚、咖啡因和马来酸氯苯那敏的含量 被引量:21

Determination of Acetaminophen,Caffeine and Chlorphenamine Maleate in Ganmaolin Granules by HPLC

摘要 目的 建立高效液相色谱法测定感冒灵颗粒中对乙酰氨基酚、咖啡因和马来酸氯苯那敏3种化学药物的含量。方法采用色谱柱Thermoelectroncorporationhypersilgold C18(4.6mm×150mm,5μm),以乙腈.0.5%磷酸(用三乙胺调至pH2.1)(6:94)为流动相,流速1.0mL·min^-1,检测波长264/249nm。结果对乙酰氨基酚、咖啡因和马来酸氯苯那敏线性范围分别为0.3245~1.1357μg(r=0.9999),0.1923~0.6731μg(r=0.9999),0.1754~0.6140μg(r=0.9998);平均回收率(n=9)分别为99.42%(BSD=0.76%)、98.58%(RSD=1.89%),98.58%(RSD=1.30%)。结论该方法准确、灵敏、重复性好,可同时测定感冒灵颗粒中3种化学药物的含量。 OBJECTIVE To develop a HPLC method for determining the contents of acetaminophen, caffeine and chlorphenamine maleate in Ganmaolin Granules. METHODS The assay was conducted on the hypersil gold C,s column(4. 6 mm× 150 mm,5 μm) of Thermo electron corporation with the mobile phase of acetonitrile-0. 5 % phosphoric acid (with triethylamine to pH 2. 1 ) (6:94 ) at the 1.0 mL · min^-1 flow rate and the detection wavelengths were 249 and 264 nm. RESULTS The linear calibration curves of acetaminophen,caffeine and chlorphenamine maleate were obtained in the ranges of 0. 324 5 - 1. 135 7 μg ( r = 0. 999 9 ) ,0. 192 3 - 0. 673 1 μg (r =0. 999 9) and 0. 175 4 -0. 614 0 μg (r =0. 999 8), respectively. Their recoveries (n =9) for the assay were at 99.42% ( RSD = 0. 76% ), 98. 58% ( RSD = 1.89% ) and 98. 58% ( RSD = 1.30% ) respectively. CONCLUSION The HPLC assay can be applied to control the product quality of the granules with a good accuracy, sensitivity and reproducible.
