摘要
采用铁盐催化比色法测定从葡萄籽中提取的原花青素含量,考察了反应时间、反应温度、反应介质的种类、酸的种类、酸的量、金属离子的种类及Fe3+的含量等因素对显色反应的影响,确定最佳测定条件为:以正丁醇及5 mL的浓盐酸的混合液作为反应介质,0.415 mol/L的Fe3+作为催化剂,60℃反应60 min的条件下,吸光度最大,体系中的原花青素酸解产生的花色素量最大,该方法准确度高,重现性好.
The proanthocyanidin content from grape seeds was determined by colormetry using ferric ions as catalyst. Effects of reaction time, reaction temperature, reaction media, acids, the concentration of acid, ferric ions concentration, metal ions, ect. on the chromogenic reaction were studied. The optimum conditions of determination were as follows: mixture of n-butanol and 5 mL hydrochloric acid as reaction medium, 0. 415 mol/L Fe^3+ as a catalyst, reaction temperature 60 ℃, and reaction time 60 min. Under the above conditions, the largest absorbance was obtained, and the quantity of eyanidin produced by acid hydrolysis of proanthoeyanidins was maximum.
