摘要 目的研究PTEN基因表达对乳腺癌细胞生长增殖的影响。方法将分别携带有野生型、突变型PTEN基因的真核表达载体pBP-wt-PTEN以及pBP-G129R-PTEN,以脂质体介导法转入人PTEN表达缺失的乳腺癌细胞ZR-75-1中,嘌呤霉素筛选获得稳定表达PTEN基因的转染细胞,通过细胞活力实验、流式细胞术观察PTEN基因对ZR-75-1细胞生长增殖的影响。结果稳定转染PTEN基因的ZR-75-1细胞中不仅PTEN蛋白稳定表达,而且其生长速度较对照组明显减慢。流式细胞术显示,细胞周期发生G1期阻滞。结论外源性PTEN基因导入ZR-75-1细胞后,可引起ZR-75-1细胞生长阻滞,抑制肿瘤细胞增殖。 Objective To investigate the effect of PTEN on cell proliferation in human breast cancer cell. Methods The eukaryotic expression vectors pBP- wt- PTEN (containing a wild -type PTEN gene) and pBP- G129R -PTEN (containing a mutant PTEN gene) were used to transfect the PTEN -null breast cancer ZR -75 - 1 cells by lipofectamine. After puromycin selection, ZR -75 - 1 cells which stably expressed PTEN were obtained and amplified. The effects of PTEN gene on cell proliferation were observed by the blue exclusion assay and flow cytometry. Results There were significanctly high expression of PTEN mRNA and protein in the transfected ZR -75 - 1 cells, whose growth rate was significantly slower than that of control. Flow cytometry showed that the progression from the G1 to S phase was arrested in cells expressing wild - type PTEN. Conclusion When transfected into ZR - 75 - 1 cells, exogenous PTEN can cause the ceil growth arrest to G1, and inhibit cell proliferation.
