摘要
将40只20日龄雌性昆明系小鼠随机分成3个处理组和1个对照组,每组10只。给3个处理组小鼠分别连续2d腹腔按体质量注射醋酸铅10,20,40mg/kg,对照组小鼠注射等体积的生理盐水。于注射后24,72h分离卵巢,用原位末端标记法(TUNEL)测定卵巢颗粒细胞的凋亡率,研究卵巢组织结构及卵巢颗粒细胞凋亡的变化。结果显示,醋酸铅可使小鼠卵巢组织结构发生病变,加速卵巢颗粒细胞的凋亡,且凋亡率随着攻毒剂量的增加和时间的延长而升高,与对照组相比,差异极显著(P<0.01);表明醋酸铅对小鼠卵巢具有毒性作用,可诱导卵巢颗粒细胞发生凋亡,并呈现一定的剂量-时间依赖关系。
Forty 20-day-old female Kunming mice were divided randomly into 3 treatment groups and 1 control group with 10 of each groups. The 3 treatment groups were injected abdominaly with 10,20 and 40 mg of lead acetate per kilogram of body weight for 2 days respectively,and the control group injected with the same volume of normal saline as the 3 treatment groups. The apoptosis rates of granulosa cells were detected by the TdT-mediated dUTP-biotin nick end labeling (TUNEL) and preparation of paraffin section on days 1 and 3 post-injection respectively,in order to investigate the change of lead acetate on the ovarian structure and apoptosis of granutose cells mice. The results indicated that the normal ovarian structure was changed throught effect of lead acetate, meanwhile the apoptosis of granutosa cells were accelerated, and the apoptosis rates of ovarian granulosa cells in every poisoning group increased significantly(P〈0. 01)with relation to the increase of lead acetate dosage and time, compared with the control group. It is concluded that the lead acetate is harmful to ovarian,inducing the apoptosis of mouse ovarian granulosa cells with being dependent on the dose and time.
