天麻对帕金森病模型鼠肿瘤坏死因子α及胶质源性神经营养因子表达的影响 被引量:3

摘要 目的探讨天麻对帕金森病模型鼠黑质(SN)和腹侧被盖区(VTA)TNF-α及胶质源性神经营养因子(GDNF)表达的影响。方法 Wistar大鼠60只,随机分为正常组、模型组、美多巴组与大、中、小剂量天麻组,每组10只,采用免疫组织化学ABC法分别进行TNF-α和GDNF阳性细胞表达的测定。结果与正常组比较,大剂量天麻组和模型组左侧SN、VTA中TNF-α表达明显升高(P<0.05,P<0.01);除小剂量天麻组左侧SN、VTA中GDNF表达明显升高(P<0.05),其余各组GNDF表达差异无统计学意义(P>0.05);与模型组比较,美多巴组、中、小剂量天麻组左侧SN中TNF-α表达明显降低,小剂量天麻组左侧VTA中TNF-α表达明显降低(P<0.05),中、小剂量天麻组左侧SN、VTA中GDNF表达明显升高(P<0.05);与美多巴组比较,小剂量天麻组VTA中的GDNF表达亦明显升高(P<0.05)。结论在6-羟基多巴胺毁损纹状体模型中,小剂量天麻显著下调TNF-α的表达和上调GDNF的表达,可能是天麻神经免疫调节的机制之一。 Objective To reveal the mechanism of the effect of gastrodin on the expression of TNF-α and gila-derived neurotrophic factor(GDNF) of Parkinsonism rat. Methods The Wistar rats were divided into 6 groups at random:normal group, 6-OHDA model group,madopa group, big,middle and small dose gastrodin groups. The expression of TNF-a and GDNF was evaluated by immunohistochemical ABC method. Results Compared with normal group,in big dose gastrodin group and 6-OHDA model group,TNF-α significantly increased (P 〈 0.05,P 〈 0.01). Compared with 6-OHDA model group,in madopa group and middle dose gastrodin group, the expression of TNF-α in substantia nigra(SN) significantly attenuated. Small dose of gastrodin significantly attenuated the expression of TNF-α in VTA (P〈 0.05) ,middle and small dose of gastrodin significantly increased the expression of GDNF in SN and VTA (P 〈 0.05). Compared with madopa group,small dose of gastrodin significantly increased the expression of GDNF in VTA. Conclusion In 6-OHDA-injured striatum model,small dose of gastrodin treatment not only significantly attenuates the expression of TNF-α, but also up-regulates the expression of GDNF. This may be one of the mechanisms of neuro immunological regulating action of gastrodin.
