利用Nestin筛选分离培养大鼠口腔黏膜固有层前体细胞 被引量:1

摘要 目的:对大鼠口腔黏膜固有层中前体细胞进行分离培养。方法:以nestin为标志物,利用免疫磁珠间接筛选法,对大鼠口腔黏膜固有层前体细胞进行筛选;免疫荧光鉴定,并用MTT法、流式细胞术检测其特性。结果:筛选细胞在体外呈克隆性生长,免疫荧光显示表达nestin、波形丝蛋白,不表达角蛋白;增殖活力旺盛;流式细胞检测结果显示85.02%的细胞处于G0/G1期。结论:成功分离培养出大鼠口腔黏膜固有层的间充质源性前体细胞,可为组织工程组织构建提供种子细胞。 Objective: To isolate progenitor cells from rat oral mucosa lamina propria and expand them in vitro. Methods: Using nestin as the marker, cells were isolated from rat oral mucosal lamina propria (OMLPPC) by immunomagnetic sorting technique. The selected cells were harvested and expanded in vitro. Then the cells were identified by immunofluorescence assay. The proliferation activity of the selected cells was tested by MTT. Furthermore, the cell cycle was analyzed by flow cytometer (FCM). Results: The selected cells behaved like clonogenic cells and showed high activity of proliferation in vitro. For these cells, nestin and vimentin were positive, cytokeratin was negative. The result of FCM showed that 85.02% of cells lied in G0/G1 period of cell cycle. Conclusion: It is demonstrated that OMLPPC is a population of mesenchymal progenitor which exists in rat oral mucosal lamina propria. Oral mucosa lamina propria may be a source of mesenchymal progenitors, and supplies seed cells for tissue engineering.
