摘要
本试验建立了用高效液相色谱法同时测定淡水鱼肝脏中的VA1和VA2的方法。以μ-Porasil3.9mm×150mm(WatersCo.)为色谱柱,正己烷:乙醚(87:13)为流动相,紫外350um,荧光Ex325um,Em480um双道检测,VA1和VA2的保留时间分别为2625min和28.00min。采用不同波长条件下的紫外吸收特性,对VA2予以定性,以VA1为内标物对VA2予以定量。实验对多种淡水鱼肝脏中的VA1和VA2进行了测定,取得了满意效果。
The high performance liquid chromatography method for a simultaneous deter-ndnation of vitamin A1 and vitamin A2 in various kinds of fresh water fish livers has been developed. Seperation was achieved by use of μ-Porasil column (3. 9 mm ×150 mm), n-hexane: diethyl ester (87: 13) as mobile phase and was detected at UV 350 nm. The qualitative assay of vitamin A1 was determined with UV absorption character in different wavelength condition and the quantitative assay of vitamin A2 were determined with vita-min A1 standard as internal standard. The vitamin A1 and vitamin A2 contents in various kinds of fresh water fish livers were determined satisfactorily.
