摘要
目的:探讨加减抵当汤对IR大鼠血浆t-PA、PAI-1水平及脂肪PAI-1mRNA表达的影响。方法:造模大鼠随机分成正常组、模型组、加减抵当汤高、中、低剂量组、文迪雅组,用发色底物法检测t-PA、PAI-1水平;RT-PCR方法检测加减抵当汤对IR大鼠脂肪PAI-1 mRNA表达的影响。结果:加减抵当汤高、中剂量组能明显升高IR大鼠血浆t-PA水平,降低PAI-l水平(P<0.05和P<0.01),下调PAI-1mRNA的表达。结论:加减抵当汤能升高血浆t-PA,降低PAI-1;并能下调脂肪PAI-1mRNA的表达,可能是其改善IR的机理之一。
Objective: To explore the effect of modified Didang decoction on t - PA, PAl - 1 and PAl - 1 gene expression in fat in insulin resistance rats. Methods:The insulin resistance rats were randomly divided into the control group, the model control group, the high, medium and low dosage of modified Didang decoction groups and Wendiya group. Chromogenie Assay kit and Reverse transcriptase polymerase chain reaction were used to observe the effect of modified Didang decoction on the t - PA, PAl - 1 and PM - 1 mRNA of fat in rats. Results : The t - PA was significantly higher and PAl - 1 and PAl - 1 gene expression level significantly lower in the high and medium dose of modified Didang decoction ( P 〈 0.05 or P 〈 0.01 ). Conclusion : Through up - regulation on t - PA and down - regulation on PAI - 1 and PAI- 1 gene expression is possibly one of the mechanisms of modified Didang decoction in treating insulin resistance.
