摘要
目的研究用表皮生长因子受体(EGFR)反义寡脱氧核苷酸(ASODN)抑制人大肠癌细胞株HR8348的生长增殖,探讨大肠癌治疗的新途径.方法人工合成的EGFR反义ODN及无关对照ODN经脂质体包裹后作用HR8348细胞,采用MTT法,[3H]TdR掺入试验,细胞周期分析,裸鼠体内接种等方法测定细胞体内外增殖的变化.结果经EGFR反义ODN处理的HR8348细胞与对照组HR8348细胞相比,体外增殖速度减慢(细胞增殖抑制率达71%)、DNA合成受抑(细胞增殖指数为0361vs0784,S期细胞数为348%vs599%)、裸鼠体内成瘤率下降(25%vs100%).结论EGFR反义ODN可有效抑制大肠癌细胞的体内外生长增殖。
IM Abstract:To investigate the effect of antisense EGFR oligodeoxynucleotides (ODNs) on human rectal carcinoma cell line HR8348.METHODS Acationic liposome and lipofectin were selected as a delivery vectoe. HR8348 cells were treated with antisense ODNs in vitro. HR8348 cells with or without pretreatment were inoculated into nude mice.RESULTS HR8348 treated with antisense EGFR greatly inhibited the proliferation with a rate of 71% and DNA synthesis (PI of EGFR group vs control group, 0361 vs 0784) of HR8348 cell line, decreased coloning formation in anchorageindependent assay. In vivo tumorigenic rate was greatly reduced in athymic nude mice as compared with untreated cells group (25% vs 100%).CONCLUSION Antisense EGFR ODNs could inhibit the proliferation of human colon cancer cell line in vitro as well as in vivo, the results implicate the potential value in colorectal cancer gene therapy.
