洪山鸡提纯选育效果的微卫星分子标记分析

摘要 利用8个微卫星座位对洪山鸡的提纯选育效果进行分析,分别计算了群体有效等位基因数、观测杂合度、期望杂合度、多态信息含量、遗传相似系数和近交系数等相关参数。结果表明:在零世代基础群和五世代洪山鸡群体中共检测到129个等位基因,片段长度在120-300bp之间;2个世代的遗传多样性指标大多表现为高度多态,零世代基础群和五世代平均多态信息含量分别为0.8614和0.6499,平均观察杂合度分别为0.4900和0.3438,平均期望杂合度分别为0.8662和0.8265;五世代的群体平均遗传相似系数为0.7759,较零世代基础群体有显著提高(0.6730);2个世代间的近交系数均值为0.0477。 Eight microsatellites locus were used for analyzing the breeding selection results of Hongshan chicken. The effective number of alleles(Ne), observed(Ho) and expected(He) heterozygosity, polymorphism information content(PIC),genetic similarity(Gs) and inbreeding coefficients(FST) were all calculated in different populations. The results from 120 to 300 base pairs,and 129 alleles were showed that all the PCR products lengths were varied detected in the maternity population and the fifth generation of Hongshan chicken. The eight locus were mostly high polymorphic. The average PIC was 0. 861 4 and 0. 649 9, the average observed and expected heterozygosity were 0. 490 0,0. 886 2 and 0. 343 8, 0. 826 5 in the two generations,respectively. The average coefficient of genetic similarity was 0. 775 9 in the fifth generation and it was significantly higher than the maternity population(0. 673 0). The average inbreeding coefficient of the two generations was 0. 047 7.
