摘要
微生物天然产物分离纯化是获得天然产物活性化合物的重要环节。以分离放线菌菌株WS-13182发酵提取物中活性物质为研究对象,采用制备液相色谱-梯度洗脱法,考察了不同色谱柱和流动相配比对样品分离度的影响,建立了适宜的分析、制备分离条件:采用美国SunfireC18柱(150*2.1mm(i.d)),粒径3.5μm,流速为0.3mL/min,流动相为乙腈和水,加入0.2%乙酸(体积比),梯度洗脱,乙腈的浓度在25min内由40%变至100%(体积比)。将该分析条件放大到制备液相色谱上,从放线菌菌株WS-13182的发酵提取物中制备得到8个化合物,其中6个化合物纯度达90%以上,经LC-MS鉴定为同系物。
Separation and purification of microbial natural products is essential for obtaining natural products. The effects of different chromatographic columns and compositions of mobile phase on resolution of samples were studied by preparative liquid chromatography-gradient elution method, and separating bioactive metabolites from fermentation extracts of actinomycetes WS-13182 as experimental material. Then, the suitable conditions for analysis and preparative separation were established as follows: using American Sunfire C18 column (150*2.1mm(i.d)), particle dia- meter was 3.5 μm, the flow velocity was 0.3 mL/min, the mobile phase was acetonitrile and water with 0.2% acetic acid (the volume ratio), gradient elution, the concentration of acetonitrilc was risen from 40% to 100% within 25 minutes (the volume ratio). To scale up the conditions on preparative liquid chromatography, then, eight compounds were obtained from fermentation extracts of actinomycetes WS-13182, of which six were in high purity (over 90%). The LC-MS analysis showed that the isolated compounds were homologue.
