摘要
目的检测人非小细胞肺癌(NSCLC)组织中Livin和Caspase-9蛋白的表达情况,并进行相关性分析。方法采用免疫组化SP法分别检测58例NSCLC组织、50例癌旁肺组织、8例正常肺组织中Livin和Caspase-9蛋白的表达水平,并分别分析两种蛋白表达与NSCLC的组织类型、分化程度、临床分期和淋巴结转移之间的关系,以及两种蛋白在NSCLC中表达的相关性。结果NSCLC组织中Livin蛋白的阳性表达率(65.5%)高于癌旁肺组织(26.0%)和正常肺组织(0%),差异有统计学意义(P<0.05);Livin蛋白在腺癌中的阳性表达率(87.5%)明显高于鳞癌(46.5%)及腺鳞癌(66.7%),在有淋巴结转移的NSCLC中的阳性率(83.3%)明显高于无淋巴结转移者(46.4%),差异均有统计学意义(P<0.05);而在不同分化程度及临床分期的NSCLC中的阳性率差异无统计学意义(P>0.05)。NSCLC组织中Caspase-9蛋白的阳性表达率(31.0%)低于癌旁肺组织(82.0%)和正常肺组织(87.5%)(P<0.05);有淋巴结转移的NSCLC组织中Caspase-9蛋白的阳性表达率(16.7%)低于无淋巴结转移者(46.4%)(P<0.05);在不同组织学类型、分化程度、临床分期的NSCLC中的阳性表达率差异无统计学意义(P>0.05)。两种蛋白在NSCLC组织中的表达呈负相关(R2=0.1447,P=0.003)。结论NSCLC组织中Livin蛋白表达增高,并与Caspase-9蛋白的表达呈负相关。
Objective To detect the expression of Livin and Caspase-9 proteins in non-small cell lung carcinoma (NSCLC) tissues and analyze the correlation between them. Methods The expression of Livin and Caspase-9 proteins were detected by streptavidin peroxides immunohistochemistry method in 58 cases of NSCLC ,50 pericarcinomatous tissues and 8 normal lung tissues. Expression of these proteins were analyzed for correlation with histological typing, degree of differentiation, clinical stage and lymph node metastasis. Results The positive expression rate of Livin protein in NSCLC (65.5%) was higher than those in pericarcinomatous lung tissue (26.0%) and normal lung tissue (0%) (P 〈 0. 05 ). The positive expression rate of Livin protein in NSCLC tissue was related with histological typing and lymph node metastasis ( P 〈 0. 05 ), except for differentiation degree and clinical stage ( P 〉 0. 05). Caspase-9 protein showed lower expression in cancers ( 31.0% ) than non-cancerous tissues ( 82.5%, 87.5%, ( P 〈 0. 05 ). The positive expression rate of Caspase-9 protein in NSCLC tissue was related with lymph node metastasis (P 〈 0.05 ), except for differentiation degree, clinical stage and histological type ( P 〉 0. 05 ). The expression of Livin protein was negatively related to that of Caspase-9 protein in NSCLC tissues (R2 = 0. 1447, P = 0. 003 ). Conclusion Over-expression of Livin and Lower-expression of Caspase-9 proteins can be seen in NSCLC, the two proteins may be negatively related with eath other.
