摘要
目的观察柴胡皂甙D(SSD)对SMMC-7721肝癌细胞的体外分化诱导作用,并初步探讨其作用机制。方法通过观察细胞毒作用、平均核面积、细胞周期和P38MAPK、NF-κB信号转导分子的表达变化初步揭示其作用机制。结果SSD具有明显的增殖抑制作用且呈一定的量效关系;SSD 2.5 mg/L、5 mg/L处理组能明显缩小平均核面积(P<0.05);能使细胞周期阻滞于G0/G1期(P<0.005);并明显上调P38MAPK表达(P<0.05),显著抑制NF-κB的核内表达(P<0.05)。结论SSD对SMMC-7721细胞具有显著诱导分化作用;并且将SMMC-7721的细胞周期明显阻滞于G0/G1期,其机制可能与上调P38MAPK,下调NF-κB表达有关。
Objective To study if Saikosapollin -d could induce differentiation, and to discuss the mechanism of action. Methods By observing effects of SSD on the average area of nucleus, eytotoxie action, cell cycle, P38MAPK and NF - κB expression in hepatocellular line SMMC -7721, the mechanism of differentiation was preliminarily revealed. Results SSD had significant cyto - inhibition and showed a doredependent response ; The average area of cell nucleus diminished at the SSD concentration of 2.5 mg/L and 5 mg/L (P 〈 0.05, vs control ) ; SSD could obviously hold - up the cell cycle in G0/G1 ( P 〈 0.05 ) by up - regulating the expression of P38MAPK( P 〈 0.05 ) and down - regulating the expression of NF - KB ( P 〈 0.05 ). Conclusion SSD induce differentiation of SMMC - 7721 cell. SSD can hold - up the cell cycle in G0/G1, probably by up - regulating P38MAPK and down regulating NF - κB.
