摘要
目的对阿片类成瘾觅药行为的药物干预的可能性进行探索。方法将40只雄性昆明种小鼠随机分为盐水组、吗啡组(9mg/kg,皮下注射)、舒必利(20mg/kg,皮下注射)+吗啡组、舒必利(30mg/kg,皮下注射)+吗啡组,每组10只,观察多巴胺D2受体拮抗剂舒必利对吗啡所致小鼠条件性位置偏爱的效应。结果吗啡组在相应的箱体中停留的时间较对侧显著延长(P<0.01)。盐水组、舒必利20mg/kg+吗啡组、舒必利30mg/kg+吗啡组与吗啡组之间比较,显示吗啡组在其相应的箱体中停留的时间显著增加(P<0.05),而其它组无显著增加。结论提示预先给予舒必利能消除吗啡产生的条件性位置偏爱。
Objective The goal of this study was to explore the possibility of the dopamine D 2 antagonists sulpiride in treating of the drug seeking behavior in mice with morphine addiction. Methods Forty mice were assigned into four groups randomly and received different treatments as following: saline S.C.; morphine 9 mg S.C.; sulpiride 20 mg/kg + morphine 9 mg/kg S.C. and sulpiride 30 mg/kg + morphine 9 mg/kg S.C.. The effect of sulpiride as a dopamine D 2 receptor antagonist agent on morphine conditioned place preference in mice was observed. Results The morphine (9 mg/kg, S.C.) group showed significantly increase the time spending in the morphine paired side during the test session relative to the opposite side ( P <0.01). It was showed that the mice in the morphine group spend a significantly longer time in the morphine paired side ( P <0.05), but not in the other groups. Conclusion These results suggest that pretreatment with sulpiride eliminates the development of morphine conditioned place preference.
