降低沙棘组培过程中外植体褐化的研究 被引量:5

摘要 为了解沙棘组培过程中影响外植体褐变的相关因子,本试验通过对沙棘组织培养中外植体取材时间、品种、不同的激素及浓度、以及活性炭加入的量等影响外植体褐化因子进行研究。结果表明,以春季休眠芽为外植体获得的沙棘组培苗防褐化效果较好,品种比较结果均显示橙色褐化较重,阿列伊,向阳,优胜褐化较轻,用l/2MS+BA3.0mg·L-1+IAA0.5mg·L-1+活性炭1.5g·L-1的培养基培养可以降低褐化率,使沙棘组培获得成功。 This experiment studies factors affecting explant browning in the sea-buckthorn tissue culture,such as drawing materials time of sampling, varieties of sampling, the different concentrations of different hormones, and the amount of the activated carbon joined. The results showed that the best anti-browning plantlet of sea-buckthorn was dormant buds in spring, the comparative varieties results showed that the orange browning was serious, the others browning were lighter.To reduce tissue browning, we can use the medium including 1/2MS and 3.0 mg'L^-1 BA and 0.5 mg'^-1 IAA and 1.5 g'L^-1 activated carbon. It was favorable for the growth of sea-buckthorn.
