重度联合免疫缺陷(SCID)小鼠排卵、胚胎发育能力及妊娠母体激素动态 被引量:3

Ovulation and Embryonic Development and Changes in Plasma Concentrations of Luteinizing Hormone and Progesterone during Pregnancy in SCID Mouse

摘要 为探讨先天性T-B淋巴细胞联合缺陷对生殖功能的影响,用正常BALB/c小鼠作对照,对重度联合免疫缺陷(SCID)小鼠的排卵、胚胎(胎儿)发育能力及妊娠母体外周血浆促黄体素(LH)和孕酮(P)水平的变化进行了研究。首先,观察了妊娠期、窝产仔数,发现SCID小鼠大多在妊娠第20天(检栓当天为第1天)凌晨分娩,平均窝产仔数为(3.86±1.67)只(n=42),BALB/c小鼠多在妊娠第20天较晚或第21天凌晨分娩,平均窝产仔数为(4.44±1.54)只(n=20),比SCID小鼠约多0.6只。取10周龄SCID雌鼠40只,BALB/c雌鼠50只,分别于第2、6、10、15、19(SCID)或20(BALB/c)天早晨各取5~10只用于试验。结果表明:(1)SCID小鼠平均排卵数比BALB/c小鼠少约0.7个(分别为8.00±1.90和8.66±2.26),而产前1d活胎数占排卵数的百分比在SCID小鼠和BALB/c小鼠间无差异(分别为66.47%和66.38%),说明SCID小鼠产仔数比BALB/c小鼠产仔数低是由于排卵数少造成的;(2)SCID小鼠在妊娠最后5d,外周血浆水平下降迅速,这期间胎儿死亡率高;BA? The effects of severe combined imunodeficiency on reproduction have been determined in the present experiments by studying ovulation, embryonic development and changes in plasma concentrations of LH and P during pregnancy in SCID mice. First of all, we have observed the length of gestation and litter size and found that most of the pregnant SCID mice delivered before dawns on d20(the day the plug was found was designated d1 of pregnancy) with a litter size of 3.68±1.67,whereas the BALB/c mice gave birth to (4.44±1.54)babies in the late on d20 or before dawn on d21 of pregnancy. Forty to fifty of 10 week old female SCID and BALB/c mice respectively were used in the following experiments. Pregnant mice on d2, d6, d10, d15 and d19(SCID) or d20 (BALB/c) were exsanguinated from the intra orbital sinus and then killed and embryos/fetuses were examined. Concentrations of plasma LH and P were assayed. Results showed that: 1) ovulation rate of SCID and BALB/c mice were 8.00±1.90 and 8.66±2.26 respectively, the difference was about 0.7 which equals approximately to the difference of litter size (0.6), while the percentage of live fetuses to ovulated ova(counted by number of corpus lutea) on one day before parturition were the same(66.47% and 66.38%, respectively), suggesting that the difference of litter size was caused by the difference of ovulation rate.2) fetal mortality in SCID mice was the highest during the last 5 days of pregnancy when concentration of P decreased rapidly. But fetal mortality in BALB/c was the highest and concentration of P decreased greatly during d10 to d15.3) There was no significant difference in concentrations of LH during pregnancy between SCID and BALB/c mice, however, all of the highest level(d15, (61.01±25.25)nmol/L), the lowest level (d2, (5.84±4.84)nmol/L) and average level during pregnancy (( 31.69 ± 22.24 )nmol/L) of P in SCID mice were lower than those in BALB/c mice (d10, (79.80±27.90)nmol/L; d2, (13.69±9.92)nmol/L and (44.75±21.46)nmol/L, respectivly). Thus we conclude that severe combined immunodeficiency affects ovarian events related to ovulation and steroid secretion in mouse.
