摘要
目的:观察扶芳藤对兔心肌缺血再灌注损伤诱发心肌细胞凋亡的保护作用。方法:40只新西兰大白兔随机分为5组,每组8只。假手术组(A组):开胸,前降支穿线不结扎。缺血再灌注组(I/R)(B组):结扎前降支40min后剪线松开行再灌注40min。扶芳藤大剂量组(C组):再灌注前5min给予扶方藤3.6g/(kg.d),余同I/R组。扶芳藤中剂量组(D组):再灌注前5min给予扶方藤1.8g/(kg.d),余同I/R组。扶芳藤小剂量组(E组):再灌注前5min给予扶方藤0.9g/(kg.d),余同I/R组。以心肌凋亡细胞原位标记法(TUNEL)观察细胞凋亡的程度,以链霉菌蛋白-过氧化物酶(SP)免疫组织化学法观察各组Bcl-2、Bax蛋白表达。结果:I/R组凋亡心肌细胞明显增加,凋亡细胞阳性指数高。扶芳藤组凋亡细胞呈散在分布,凋亡细胞阳性指数较I/R组低,而且剂量越高,凋亡细胞阳性指数越低(P<0.01)。再灌注后I/R组Bcl-2和Bax的表达量均有所上升,但以Bax蛋白表达更明显。与I/R组比较,扶芳藤组Bcl-2表达显著增高,而Bax蛋白显著降低,尤以大剂量组明显(P<0.01)。结论:扶方藤可以减少Bax蛋白的表达,升高Bcl-2,从而抑制再灌注心肌细胞凋亡。
Objective:To observe the protective effect of Euonymus fortunei Zhuang drugs on rabbit myocardial ischemia - reperfusion injury - induced cardiomyocyte apoptosis. Methods : A total of 40 New Zealand white rabbits were randomLy divided into 5 groups of 8. Sham - operated group ( Group A) : Thoracotomy, anterior descending artery threading without ligation. Ischemia - rcperfusion group(I/R) (Group B) :40min anterior descending artery ligation, then shear line release and 40 min reperfusion. High-dose group of Euonymus fortunei( Group C ) : Give the oral Euonymus fortunei 3.6g/( kg · d) 5 minutes before the reperfusion,then to do the same performances as the group(I/R). Mid-dose group of Euonymus fortunei (Group D) :Give the oral Euonymus fortunei 1.8g/(kg · d) 5 minutes before the reperfusion,then to do the same performances as the group(I/R). Low-dose group of Euonymus fortunei (Group E) :Give the oral Euonymus fortunei 0. 9g/( kg · d) 5 minutes before the reperfusion, then to do the same performances as the group (I/R). To observe the extent of apoptosis and Bcl - 2, Bax protein expression with methods of Myocardial apoptotic cells in situ labeling(TUNEL) and Protein streptavidin-peroxidase(SP) immunohistochemical. Results:I、R Group's significant increase in cardiac myocyte apoptosis ,apoptotie cell -positive index is high ,And the higher dose,apoptotie cells were lower index( P 〈 0. 01 ). After reperfusion,i/R group's Bcl - 2 and Bax expressions increased in both, but more obviously is Bax protein expression. Compare with the I/R Group, Euonymus fortunei group's Bcl -2 expression was obviously higher, and a obvious reduction in Bax protein, particularly the results of high - dose group ( P 〈 0.01 ). Conclusion : The Euonymus fortunei can help to inhibit cardiomyocyte apoptosis of myocardial reperfusion by reducing Bax protein expression and increasing Bcl - 2.
