摘要
研究钉螺谷草转氨酶(GOT)的催化最适条件及对Niclosamide(NiC,氯硝柳胺)浸泡染毒的敏感性。取钉螺软体组织匀浆,将13000r/min离心的上清液作为GOT酶液,用L25(56)正交设计和极(方)差分析GOT活性测定最适条件:酶液质量浓度0.2g/L,底物浓度5mmol/L,反应体系pH值7.5,反应温度45℃,反应时间60min。与去氯水对照组相比较,1.5mg/LNiC浸泡钉螺24、48h,其软体GOT活性抑制率分别达25.98%、26.81%。结果说明GOT可能是NiC胁迫的防御性靶酶之一。
The optimal catalytic conditions of glutamic-oxalacetic transaminase (GOT) and its sensitivity to niclosamide (NiC) was studied. The homogenate of Oncomelania hupensis soft tissue was centrifugal by 13000 r/min and the supernatant was used as enzyme. The optimal catalytic conditions of GOT was established by the L25(56) orthogonal matrix method and range analysis: enzyme concentration was 0.2 g/L, substrate concentration was 5 mmol/L, pH of reaction system was 7.5, reaction temperature was 45℃ and reaction time was 60 rain. Compared with control group, after Oncomelania hupensis was immerged in 1.5 mg/L NiC for 24 and 48 h, the inhibition ratio of GOT activity was 25.98% and 26.81%, respectively. The results showed that GOT was probably one of target enzyme that menacingly defense to NiC.
