摘要
目的:观察肥胖哮喘大鼠体内脂联素表达水平的变化及氨茶碱对脂联素表达及气道炎症的影响。方法:将大鼠随机分为正常体重对照组(A组)、哮喘组(B组)、茶碱干预组(C组)和肥胖对照组(D组)、哮喘组(E组)、茶碱干预组(F组),建立肥胖、哮喘模型。计数支气管肺泡灌洗液(BALF)细胞总数,图像分析软件测定气道壁厚度,ELISA测定血清、BALF脂联素浓度,RT-PCR测定脂肪组织脂联素mRNA的表达。结果:(1)BALF细胞总数和支气管壁厚度,E组较B组均有所升高,C、F组分别较B、E组明显降低。(2)脂肪组织脂联素mRNA表达及血清、BALF脂联素浓度,E组较B组明显降低,C、F组分别较B、E组明显升高。结论:脂联素表达显著降低是肥胖哮喘大鼠气道炎症更为明显的重要原因。氨茶碱能抑制气道炎症并部分恢复哮喘大鼠脂联素表达。
Objective To observe the expression of adiponectin in obese rats with asthma and to explore the effects of aminophylline on it and airway inflammation. Methods Rat were randomly divided into 6 groups (each n= 21): 3 groups with normal weight [control group (group A), asthmatic group (group B), and aminophylline intervention group (group C)] and 3 groups with obesity [control group (group D), asthmatic group (group E), and aminophylline intervention group (group F)]. Total number of ceils in bronchoalveolar lavage fluid (BALF) were counted by cytology method, thickness of airway wall was determined by an image analysis software, the concentrations of adiponectin in serum and BALF were determined by ELISA, and the expression of adiponectin in adipose tissue was measured by RT-PCR. Results The total number of cells in BALF and the thickness of airway wall in group E were higher than those in group B, while those in group C and F were significantly lower than those in group B and E, respectively. The expression of adiponectin mRNA in adipose tissue and the concentrations of adiponectin in serum and BALF were lower in group E than those in group B, while those in group C and F were significantly higher than those in group B and E, respectively. Conclusions Significantly lower expression of adiponectin is accounted for greater airway inflammation in obese rats with asthma. Aminophylline can inhibit airway inflammation and partially restore the expression of adiponectin in asthmatic rats.
